I have already some leveled reds on my line ups:
BT 4.70+4
Colen 3.60
Gormek 3.60
Currently trying to level another red. In line, I have: 1 Scarlett, 4 Colens.
Which one do you think I should level next for my red: Scarlett or 2nd Colen?
I have no 5* red yet.
And lacking of compass to ascend either 3.60 Gormek or Colen.

Definitely Scarlett… She is the only 4* red attack debuffer and has fast mana. Also, her tile damage is extremely high so very good for titans and red colour stacking in general.

Indeed I need 4th 4* red for color stacking against green tank or green titan, to replace 3* Azar…
And when I get the compass, should I also prioritize Scarlett over Gormek?
Well, when I get the compass, I have some already in line:
3.60 Grimm
2.60 Poseidon
2.60 Musashi
2.60 Justice
3.60 Kasshrek
3.60 Gormek
3.60 Colen
It will totally depend on what other heroes you have maxed in the other colours. I wouldn’t touch the 5 stars right now, atleast not till you have atleast 2-3x 4 stars in each color maxed as they are much more faster to level and consumes less amount of materials. 5 stars achieve their full potential only when fully ascended, which might take a while. So it’s better to develop a deep bench of 4 stars before that so that you can colour stack in raids, titans, wars and challenge events.
Here is my complete roster:
For Blue, I stack: Misandra, Boril, Grimm
Green: Little John, Gadeirus, Kasshrek
Purple: Proteus, Sabina, Balthazar/Tyrum
Yellow: LiXiu, Musashi, Poseidon, Justice/Ranvir
Only Boril, BT, Little John, Sabina are maxed.
Definitely Scarlett. If you didn’t have either, I’d still say Scarlett.
I’ve found that I generally want a new hero over a duplicate hero of the same element and rarity,a s it increases my options.

In my honest opinion it would have to be Scarlett to give variety and depth

I’ve got plenty of ascended fives but still use Scarlett on Titans, insane attack stat. She is much better than Colen with her fast mana. She was better than the first Colen, let alone the second

For 2nd red 4* should it be Scarlett as well to 4th ascension - after BoldTusk? Or Gormek with his defense debuff? I have Blue Gormek as well (Grimm).
You have a pretty good roster and since you lack a blue hitter Grimm should be the next one to be ascended. And as a general rule of thumb, in the beginning it is always good to ascend a variety of heroes in each colour. Each hero has unique special skills which are really useful in certain scenarios. Once you have a deep enough roster, then you can start thinking about duplicate heroes to raise, especially the extremely good ones, for example Proteus, Wilbur, Rigard, Boldtusk, Kiril etc…
